Box-Behnken响应面法优化牛至挥发油减压辅助提取工艺及GC-MS成分分析与体外抑菌作用研究

摘要
目的:采用水蒸气减压蒸馏法提取牛至挥发油,利用Box-Behnken响应面分析法,确定了牛至挥发油的最佳提取工艺,即加水14倍,压强558mbar,加热回流4.23h,在此工艺条件下,牛至挥发油实际得率为0.9267%,与理论值较接近。并用气相色谱-质谱联用法(GC-MS)分离鉴定,比较常压和减压提取牛至精油成分的差异及所得精油对金黄色葡萄球菌和大肠杆菌的抑菌效果。结果表明,常压和减压提取所得牛至精油在成分种类和含量上都存在一定差别,抑菌实验结果显示,无论是对金黄色葡萄球菌还是大肠杆菌,减压提取所得牛至精油的抑菌效果都优于常压提取,这可能与某些抑菌成分的含量有关。
Objective Water vapor vacuum distillation method was used to extract the essential oil from Origanum vulgare L.,and Box-Behnken central composite experimental design and response surface analysis method were used to determine the optimal extracting process of volatile oil of oregano,that is,add water 14 times,pressure 558 mbar,heating reflux 4.23 h.Under these conditions,oregano essential oil actual yield was 0.9267%,and is close to the theoretical value.And composition analysis with gas chromatography-mass spectrometry(GC-MS),compare the differences of the bacteriostatic effect of oregano essential oil extracted under the condition of normal pressure and decompression.Results show that there are some differences in the type and content of the oregano essential oil composition extracted under the condition of normal pressure and decompression.And bacteriostatic experiment results show that the bacteriostatic effects of oregano essential oil extracted under the condition of reduced pressure are better than normal pressure extraction both to staphylococcus aureus and escherichia coli,this may be related to the content of certain bacteriostasis component.
