Pigs are important for agriculture and study of human diseases.Transcription activator-like effector nucleases(TALENs) and clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated(Cas) 9 system(CRISPR/Cas9) show enormous potential for gene engineering in various species,including pigs.Here,we used TALENs and CRISPR/Cas9 system to target single genes(GGTA1,Parkin,DJ-1) and triple genes(Parkin/DJ-1/PINK1) in the porcine genome.The targeting efficiency of TALENs was 2.5%-22%,while the CRISPR/Cas9 system reached an efficiency of up to 100%.DJ-1knockout(KO) piglets were generated by somatic cell nuclear transfer from targeted fibroblasts and Parkin/DJ-1/PINK1 triple targeted pigs were generated by direct embryo injection.These pigs would provide valuable information for studying Parkinson's disease in human.
