In order to meet the increasing challenge of global antibiotic resistance, a new generation of antimicrobials with broad spectrum and high efficiency must be developed. Based on previous studies and combined with data from the Antimicrobial Peptide Database, this project involves the design of a series of modified antimicrobial peptides with different compositions by a combination of structural analysis, molecular design, helical structure domain truncation and amino acid substitution with the 13 amino acid antimicrobial peptide Temporin-Pta as a template. Three antibacterial peptides, designated, 21#, 22# and 14# with good antibacterial activity were obtained by the combined technologies of microbiology and biochemistry. The main metabolic pathways and target genes of different antimicrobial peptides in anaerobic conditions were explored by comparing the antimicrobial activity of the antimicrobial peptide Temporin-Pta This work is of important scientific value for the study of the bactericidal mechanisms of different compositions of antibacterial peptides, and provides a theoretical basis for the design and development of a new type of antibacterial peptide.
