家养梅花鹿品种Y染色体遗传多样性和父系遗传结构

摘要
为了全面研究家养梅花鹿的遗传多样性及遗传结构,为家养梅花鹿育种工作提供理论依据,利用PCR直接测序法,测定了我国培育的家养梅花鹿品种和品系389头雄性梅花鹿Y染色体AMELY基因部分序列,针对测序得到的序列利用生物信息学软件进行分析,筛选出6个SNPs位点,得到了6种单倍型(H1、H2、H3、H4、H5和H6),结果表明,我国家养梅花鹿Y染色体遗传多样性较低,种群之间没有发生显著的遗传分化。单倍型H1为优势单倍型,在种群中有比较高的频率,其次为单倍型H2,其他单倍型频率比较低。
In order to investigate genetic diversity and genetic structure of domestic sika deer in China comprehensively,and provide theoretical basis for breeding of sika deer.AMELY gene fragments on Y chromosome in 389 individuals from 8 domestic sika deer populations were amplified using PCR method,and the products were sequenced directly.The sequences attained were analyzed through bioinformatics software.Six SNPs were found,and six haplotypes(H1、 H2、 H3、 H4、 H5 and H6) were observed.The domestic populations of Chinese sika deer exhibited a low level in genetic diversity on Y chromosome,and no significant genetic structure difference was found among different populations.H1 was the most dominant haplotype,showed an extraordinary high frequency across all populations.H2 showed the second frequency.Other haplotypes showed a low frequency.
