中药天七方对大鼠急性心肌缺血保护作用及机理的研究

摘要
目的:观察中药天七方对急性心肌缺血模型大鼠心肌组织的保护作用并进一步探讨其作用机制。
     方法:采用异丙肾上腺素复制大鼠急性心肌缺血模型,观察实验动物给予中药天七方后心电图改变,测定血清中磷酸肌酸激酶(CK)、天冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)及心肌组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)含量变化情况;通过ELASA法检测各组心肌缺血模型大鼠心肌组织匀浆中活性氧(ROS)、血红素加氧酶-1(HO-1)含量;通过HE染色观察各组心肌组织的病理学变化;通过Western blot法检测中药天七方对心肌缺血模型大鼠心肌组织凋亡相关蛋白p53、Bcl-2、Bax表达的影响;通过原位缺口末端标记法(TUNEL)测定细胞凋亡率。
     结果:中药天七方对于异丙肾上腺素所致急性心肌缺血模型大鼠心电图有改善作用;能够降低急性心肌缺血模型大鼠血清中CK、AST、 LDH的含量;可以增加SOD. GSH-Px含量,降低缺血所致心肌组织中MDA含量的增加,显著降低心肌组织中ROS含量,但对心肌组织中HO-1含量未见显著影响;HE染色结果显示中药天七方各剂量组心肌组织肌束排列整齐,中度炎性细胞浸润,轻度血管扩张,轻度心肌纤维化;此外通过TUNEL法测定细胞凋亡与western blot法测定凋亡调节基因表达相结合的方法显示中药天七方各剂量组均有抑制细胞凋亡的作用。
     结论:中药天七方对大鼠急性心肌缺血具有保护作用,其作用机制可能与其改善心肌酶谱、抗自由基及脂质过氧化、抑制细胞凋亡有关,而与热休克蛋白相关的机制有待进一步探讨。
Objective:To observe the protective effect of Rhodiola-Pseudo ginseng Prescription(RP Prescription) on myocardial tissue of rats with acute myocardial ischemia model,and to further investigate its mechanism of action.
     Method:Isoproterenol was used to induce acute myocardial ischemia model in this experiment. Given the RP Prescription to rats with acute myocardial ischemia model,observe the changes of electrocardiogram. Determinated changes in serum CK,AST,LDH levels,and in Myocardial tissue homogenates SOD,MDA,GSH-Px levels. By ELASA detection,Determinated changes in Myocardial tissue homogenates ROS,HO-1levels. By Western blot method,determinated effects of RP Prescription on apoptosisdetection of myocardial ischemia rats myocardial tissueassociated protein,Bcl-2,expression of Bax p53. The cell apoptosis rate was determined by TUNEL.To investigate the protective effects and mechanism of RP Prescription on myocardial tissue of rats with acute myocardial ischemia model by methods above.
     Result:RP Prescription have effect on ECG of acute myocardial ischemia model rats induced by isoproterenol. RP Prescription can reduce the content in serum of rats with acute myocardial ischemia model in CK,AST,LDH. RP Prescription decreased with the increase in MDA content induced by ischemia in cardiac tissue. RP Prescription remarkably decreased the content of ROS in myocardial tissue, but no significant effect of HO-1content in myocardium. HE staining showed that RP Prescription myocardial tissue in each dose group was arranged in neat rows, moderate infiltration of inflammatory cells, mildvascular dilatation, mild myocardial fibrosis. In addition method by combining TUNEL method and Western blot method show every RP Prescription dose groups could inhibit cell apoptosis in the model of acute myocardial ischemia rats.
     Conclusion:RP Prescription can be effective against acute myocardial ischemia in rats. The mechanism may be related to improving myocardialenzyme spectrum, anti free radical and lipid peroxidation, apoptosis. The mechanisms associated with Heat Shock Proteins (HSPs) needs to be further studied.
