纳米钙补肾中药调节骨质疏松症骨钙代谢作用机制的实验研究

摘要
本实验研究目的:通过观察纳米钙中药和补肾补钙中药复方对肾虚骨质疏松大鼠各项骨钙代谢指标及对BGP基因和蛋白表达、BMP-2基因和蛋白表达的影响,旨在从骨钙代谢方面探讨骨质疏松症的病理和中药调节机制,并观察纳米中药对钙吸收和沉积的影响。
     主要研究内容和研究方法:采用超音速气流磨纳米技术制备纳米中药。采用去势法腰背部切除双侧卵巢,建立肾虚骨质疏松大鼠模型。使用NORLAND公司XR-36型双能X线骨密度仪,采用小动物骨密度测定软件,测定大鼠左后肢股骨的骨密度;使用应变测试系统,按股骨拉伸和三点弯曲试验,测定股骨生物力学指标;以等离子体发射光谱法测定腰椎骨骨钙含量;以常规生化法测定血清钙、镁含量及血清酸性磷酸酶含量;以Elisa法测定骨钙素含量。以RT-PCR法检测大鼠右侧股骨骨钙素BGP基因和骨形成蛋白BMP-2基因表达;以Western-blot法检测右侧股骨骨钙素BGP和骨形成蛋白BMP-2蛋白表达。
     研究结果表明:大鼠去卵巢后体重明显增加、子宫重量明显降低,骨密度、骨钙含量、血清钙离子及镁离子浓度均显著降低,血清骨钙素含量显著降低,血清抗酒石酸酸性磷酸酶含量显著增高,骨生物力学指标和骨生物力学性能明显降低,BGP和BMP-2基因及其蛋白表达明显低于正常组。通过用药,纳米钙能显著提高去卵巢实验大鼠股骨的骨密度值,其效果优于阳性药对照组,也优于纳米钙补肾中药组;但在提高股骨头骨密度方面,纳米钙补肾中药稍优于纳米钙。纳米钙能显著提高去卵巢实验大鼠腰椎骨骨钙含量以及血清钙、镁含量,其效果优于阳性药对照组,也优于纳米钙补肾中药组。我们发现,通过用药能改善和恢复大鼠骨生物力学性能,但各组在治疗效果方面无明显差异。治疗组能提高去卵巢大鼠血清BGP含量和降低血清TRAP值,但治疗组和对照组均未能使二者达到正常组值。纳米钙和纳米钙补肾中药明显促进大鼠骨BGP和BMP-2基因及其蛋白表达,其中以纳米钙组表达最强,纳米钙补肾中药组其次,且呈剂量依赖性。
     结论:单味中药通过纳米细化后,能明显提高药物的吸收和在体内的沉积,提高药物疗效,其作用可能是通过影响微循环中钙离子浓度,进而影响骨细胞内外BGP和BMP-2基因和蛋白表达水平,从而促进钙的沉积和骨的矿化,抑制骨吸收,促进骨形成。
Purpose: In order to approaching the patho of the osteoporosis or the regulatory mechanism of the nanometer Calcium and the traditional Chinese medicine,observing the effects of the nanometer Calcium and the nanometer Calcium-traditional Chinese medicine complex prescription in Calcium absorptory and deposition,we observed the each indicatrix of the bone calcium metabolism and the effects of the BGP and BMP-2 from the gene and proteinum express level to the osteoporosis rat due to Kidney-Deficiency.
    Method:Using the fluidized-bed opposed jet mill and the nano technology,we prepare the nanometer Chinese traditional medicine.Using the method of ovariectomy, we established the rat model of osteoporosis due to kidney-deficiency.Using dual energy X-ray bone density instrument XR-36 model to detect the bone mineral density of the left femur of the rats. Using the strain test system, we examined the biomechanics index according the tension and three-spot bending test; we also examined the bone calcium of the lumbar vertebra through plasma emission spectroscopy . We examined the serum calcium and magnesium by using the nomal biomechanics method,and examined the serum acid phosphatase and osteocalcin by using the Elisa method. We also examined the expression of BGP and BMP-2 gene with the method of RT-PCR and the proteinum expression with the method of WESTERN-BLOT in the bone tissue of osteoporosis rats due to Kidney-Deficiency.
    Result:When the rats had been removed the ovaries,their weights of body raised and their weights of uterus depressed obviously.All of their bone density,calcium content of bone,serum calcium and magnesium ionic concentration were depressed obviously.Otherwise,the content of the serum osteocalcin was depressed and the TRAP increased obviously.The biomechanics index and property of the bone depressed obviously.The BGP and BMP-2 gene and proteinum expressiones were belower than these of the normal group.
    Through medication,Nano-Ca could increase the femoral bone density of desovary experiment rat obviously,and the effects surpassed these of masculine medicine control group, surpassed these of Nano-Ca and invigorating the kidney group. But to the head of remur, the Nano-Ca and invigorating the kidney group surpassed Nano-Ca group slightly. Nano-Ca could increase calcium content of bone of lumbar vertebra,serum calcium and magnesium,the effects surpassed these of masculine medicine control group and of Nano-Ca and invigorating the kidney group. Through medication,the biomechanics index and property of the bone were improved or recoveried, and the effects of all groups had no obviously difference.The tretment groups could improve the content of BGP and depress the content of TRAP which in the blood serum, but both of the tretment groups and control groups failed to attent the normal value. The Nano-Ca group and the Nano-Ca and invigorating the kidney group could promote the BGP and BMP-2 gene and proteinum expr
    essiones obviously. The Nano-Ca group
    
    
    expressed most strongly, and the Nano-Ca and invigorating the kidney group expressed the next, furthermore the expressing were dose dependent.
    Conclusion:When the single traditional Chinese medicine was zoomed to nanometer.the absorption and deposition of the medicine were improved obviously.The effects of treatment were improved too.The machines maybe were that the Ca-ionic concentration in microcirculation was affected,and the BGP and BMP-2 gene and proteinum expressiones in exterior and interior of osteocyte were influenced,thus the deposition of the Ca and the mineralization of the bone were improved,the bone resorption was refrained,and the bone formation was promoted.
