摘要
目的 探讨麦粒灸对痛风性关节炎小鼠滑膜细胞功能的调控及其机制。
方法 以痛风性关节炎小鼠为实验模型,分别予以麦粒灸及消炎痛治疗;以排水法进行关节肿胀程度评价;HE染色观察尿酸盐结晶及滑膜病理组织学变化;ELISA检测TNF-α、IL-2、IL-4、IL-12含量;免疫组织化学测定HSP70表达。
结果:痛风性关节炎小鼠关节肿胀、尿酸盐数目沉积增多、炎性细胞浸润、增多、关节局部TNF-α升高、IL-2、IL-12降低,艾灸后关节肿胀减轻,尿酸盐数目沉积减少,炎性细胞减少、关节局部TNF-α下降、IL-2、IL-12升高;艾灸后关节炎局部热休克蛋白(HSP 70)表达增强。
结论:麦粒灸能明显改善痛风性关节炎小鼠关节症状;其机制可能与调整关节滑膜细胞因子含量,增强关节滑膜细胞HSP70表达有关。
Objective To investigate the regulation of synovm cell function and its mechanism in gout mouse with moxibustion. Methods The gout mouse were treated respectively with indomethin and moxibustion; the swelling of joint was measured by drainage; the number of MSU and pathological feature were detected through HE colouring; The change of concentration of TNF- a ? IL-2, IL-4, IL-12 were evaluated by ELISA; the exppression of HSP70 detected by immunohistochemistry. Results: The swelling was alleviative the number of MSU was fewer after moxibustion treatment taken in gout mouse; the amount of TNF-a in joint synovm was decreased while IL-2 and IL-12 increase after 3 courses(21days) treatment with moxibustion ; the expression of HSP70 was enhanced by moxibustion. Conclusions: Wheat-moxibustion can alleviate the swelling apparently which is relation to regularting the mount of cytokine joint synovm and enhacing the expression of HSP70.
