1株耐硒凝结芽孢杆菌的分离鉴定及其培养条件的优化

摘要
用含硒培养基,采用平板稀释分离法,从湖北恩施渔塘坝高硒环境中分离鉴定1株凝结芽孢杆菌。为确定该凝结芽孢杆菌培养基及培养条件,采用单因素和正交试验法对凝结芽孢杆菌的摇瓶发酵培养条件(碳源、氮源、温度和转速)进行了优化,为确定其培养时间作出了生长曲线。结果表明,温度对凝结芽孢杆菌生长的影响最大,其后依次为氮源、碳源和转速;最适培养基为:葡萄糖11 g/L,牛肉浸膏11 g/L,氯化钠5 g/L;最适培养条件为:温度35℃,转速160 r/min;(2)从生长曲线可看出,0~1.5 h为停滞期,1.5~10 h为对数生长期,10~20 h为稳定期,最佳培养时间为10 h。
