猪肺炎支原体西昌分离株16S rRNA和P46部分基因的克隆与序列分析

摘要
为了解西昌猪肺炎支原体的遗传变异情况,对西昌市Mhp分离株XC01的16S rRNA和P46基因部分序列进行PCR扩增和序列分析,并构建NJ树。结果显示,XC01的16S rRNA和P46基因片段的长度分别为1 146 bp和1 059 bp,与Mhp参考株的核苷酸序列同源性分别为97.5%~99.8%和98.6%~99.4%;构建的NJ树显示不同地方的Mhp株未形成明显的地理分支。结果表明,Mhp分离株的16S rRNA和P46基因序列高度保守,在遗传演化中未发生太大的变化。研究结果为猪肺炎支原体的分子遗传变异研究和开展分子流行病学调查提供参考依据。
        In order to understand genetic variation of Mycoplasma hyopneumonia in Xichang city,the partial sequence of 16S rRNA gene and P46 gene of XC01 isolated from Xichang city were amplified by PCR, analyzed and constructed NJ trees. The results showed that the partial sequence of 16S rRNA gene and P46 gene of XC01 was 1 146 bp and 1 059 bp, respectively. The nucleotide sequence homology similarity was 97.5%~99.8% and 98.6%~99.4%, respectively. The NJ trees showed that different strains of M. hyopneumonia isolated from world were not obvious geography cluster.These findings indicated that sequences of 16S rRNA and P46 genes of Mhp were highly conserved,there were not much has changed in genetic evolution. The results provide reference for the study of molecular genetic variation of M. hyopneumonia and the investigation of molecular epidemiology.
