摘要
目的:通过观察TNF-a介导的NF-kb信号通路在河西走廊地区沙尘所致大鼠慢性咽炎中的作用,探讨山豆根治疗此类咽炎的作用机制。方法:将健康成年SD大鼠按随机数字表法分为4组,造模成功后脱臼处死,光镜下观察咽部组织形态学变化,并测定血白介素-1(IL-1)、白介素-6(IL-6)、肿瘤坏死因子-a(TNF-a)、丙二醛(MDA)、总抗氧化能力(T-AOC)及核转录因子-kb(NF-kb)的表达水平。结果:沙尘颗粒物可明显升高大鼠体内TNF-a、IL-1、IL-6、NF-kb、MDA的含量,降低T-AOC水平;山豆根可通过降低TNF-a等含量及提高T-AOC水平阻断沙尘对咽部组织的损伤作用。结论:山豆根治疗沙尘颗粒物所致慢性咽炎的作用机制可能是通过TNF-a受体介导的信号转导途径,下调核转录因子-kb的表达水平而发挥作用。
Objective: To observe the effect of TNF-a mediated NF-κB signaling pathway on chronic pharyngitis induced by sand and dust in Hexi Corridor area,and to explore the mechanism of action of Radix Sophorae Tonkinensis in treating such pharyngitis. Methods: SD rats were randomly divided into four groups; after successful model,the rats were sacrificed by means of dislocation and the morphological changes of the pharynx were observed under light microscope,while measuring expression levels of IL-1,IL-6,IL-( TNF-a),malondialdehyde( MDA),total antioxidant capacity( TAOC) and nuclear transcription factor-kappa B( NF-kb). Results: The contents of TNF-a,IL-1,IL-6,NF-kappa B and MDA in the rats were significantly increased by dust particles,leading to decreased T-AOC level,and the contents of TNFa were decreased due to Radix Sophorae Tonkinensis to decrease T-AOC level,blocking the role of dust on the pharyngeal tissue damage. Conclusion: The mechanism of the treatment of chronic pharyngitis caused by dust particles using Radix Sophorae Tonkinensis may play a role through the TNF-a receptor-mediated signal transduction pathway,down-regulation of nuclear transcription factor-kb expression level.
