响应面法优化白屈菜中总生物碱酸水提取的工艺

摘要
目的:优化盐酸超声提取白屈菜中总生物碱的提取工艺。方法:根据单因素试验结果,选取影响总生物碱提取率的主要因素盐酸浓度、料液体积和提取温度,以总生物碱的得率为响应值,进行响应面法中心组合试验设计,采用响应面法描述三个因素对总生物碱提取率的影响变化趋势。结果:响应面法所得模型具有显著性(P<0.000 1),白屈菜生物碱的最优提取条件为盐酸浓度0.42%,料液体积97.59 ml,提取温度50.12℃,在此条件下得到的生物碱为2.709 0 mg·g~(-1)。结论:用该方法优化提取工艺科学可靠,可用于白屈菜总生物碱的提取。
        Objective: To optimize the extraction process of total alkaloids in Chelidonium majus by hydrochloric acid ultrasound method. Methods: According to the results of single factor experiment,the concentration of hydrochloric acid,volume of feed liquid and extraction temperature were selected as the main factors affecting the extraction rate of total alkaloids. The response surface methodology (RSM) was used to design the central composite test with the total alkaloids yield as the response value,and the RSM was used to describe the changing trend of the three factors affecting the extraction rate of total alkaloids. Results: The model obtained by response surface methodology was significant (P<0.000 1). The optimum extraction conditions of alkaloids in Chelidonium majus were as follows: the hydrochloric acid concentration was 0.42%,the liquid volume was 97.59 ml,the extraction temperature was 50.12℃,and the total alkaloid obtained under the conditions was 2.709 0 mg·g~(-1). Conclusion: The optimized extraction process is scientific and reliable,and can be used for the extraction of total alkaloids in Chelidonium majus L.
