摘要
目的探讨骨髓间充质干细胞(bone marrow mesenchymalstem cells,BMSCs)移植对大鼠子宫内膜损伤的修复作用。方法 45只雌性SD大鼠分为三组:正常组、模型组、BMSCs组,每组15只。除正常组外,其余两组大鼠采用95%乙醇宫腔注射法建立子宫内膜损伤模型。正常组不予任何干预,模型组造模后注射PBS溶液,BMSCs组注射体外分离培养的雄性SD大鼠BMSCs悬液。14 d后切除子宫,观察子宫内膜形态学改变、子宫内膜厚度、用免疫组织化学方法检测子宫内膜角蛋白、波形蛋白、整合素β3的表达,通过对移植后子宫内膜组织的SRY DNA原位杂交来定位源自雄性供体的细胞。结果与正常组相比,模型组大鼠子宫内膜厚度显著变薄[(276.25±59.46)μm vs(413.52±68.67)μm,P <0.01],而BMSCs组大鼠子宫内膜的厚度[(347.22±87.56)μm]较模型组显著增加(P <0.05)。与正常组相比,模型组大鼠子宫内膜角蛋白、波形蛋白、整合素β3表达明显减少(P <0.01),而BMSCs组较模型组明显增加(P <0.05)。BMSCs组损伤侧子宫内膜组织中有SRY阳性细胞的定植,模型组则未见。结论 BMSCs移植能不同程度地促进大鼠损伤子宫内膜的修复。
        Objective To study the rapeutic effect of bone marrow mesenchymal stem cells(BMSCs) transplantation on the Rat endometrial damage. Methods 45 adult female SD rats were divided into three groups: normal control group,model control group and BMSCs group. The rat uterine endometrial damage models were established by intrauterine injection of 95% ethanol. The model controls were injected with PBS solution,the BMSCs group injected with stem cell suspension. 14 days later, the uterus were collected from the rats for observation of morphology, thickness, and the expression of cytokeratin,vimentin and integrin β3. In site hybridization was used to observe the expression of SRY-positive cells in the uterine endometrial tissue. Results Compared with the model controls,the rats of the BMSCs group showed significant thickening of the endometrium(P <0.05). The expressions of cytokeratin, vimentin and integrin β3 showed the highest in the normal control,the lowest in the model control group,and increased in theBMSCs group(P <0.05).It was shown that the BMSCs group appeared colonization of SRY-positive cells in the endometrium, while the model control group did not. Conclusion Bone marrow mesenchymal stem cell transplantation can promote the repair of damaged uterine endometrium in rats.
