单因素法考察葛仙米多糖提取工艺

摘要
为开发利用葛仙米多糖资源,基于多糖和单糖含量考察其提取工艺。通过单因素法,对工艺中提取方法、提取时间、提取温度、提取次数以及液料比进行考察。最佳提取方式为水浴提取,提取温度100℃,提取时间1.5 h,液料比200︰1 mL/g,提取3次,此条件下测得不同批次的样品中粗多糖平均得率为51.51%,水解衍生后其组成为葡萄糖醛酸、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖,单糖总量平均得率为28.72%。研究结果准确可靠,可为后续葛仙米地方质量标准中多糖提取、纯化、水解和含量测定等研究奠定基础,为规范葛仙米相关产品质量提供依据。
        For developing and utilizing the resource of Nostoc sphaeroids Kutz polysaccharides, the extraction process was investigated based on the content of crude polysaccharides and monosaccharide by single factor method. In this experiment,the process, extraction method, extraction time, extraction temperature, extraction times and liquid material ratio were investigation factors. The optimum extraction method was water bath extraction, and other extraction parameter was extracting 3 times with water(200︰1 mL/g) at 100 ℃ for 2 h. Under this condition, the coarse polysaccharide average extraction rate was51.51%. After hydrolysis-derived, the constituents of polysaccharide are glucuronic acid, arabinose, xylose, mannose, glucose and galactose. The average yield of total monosaccharide was 28.72%. The results were accurate and reliable, which could lay a foundation for the following studies on the extraction, purification, hydrolysis and content determination of polysaccharides,and also could provide a basis for the standardization of the quality of related products of Nostoc sphaeroids Kutz.
