摘要
目的建立大鼠血液中阿霉素(Dox)含量测定的新方法。方法采用荧光分光光度法测定大鼠血浆中的Dox,建立了一种快速、简便、灵敏的定量测定大鼠血浆中Dox及CS-Dox含量的方法。结果在0.1~15.0μg/ml的范围内具有良好的线性关系,回归方程为Y=16.944X+0.6796,R2=0.9997。该方法的日内精密度和日间精密度的RSD均小于2.5%,Dox及CS-Dox的提取回收率均大于84%,表明该检测方法回收率良好。结论所建方法成本低、快速、简便、灵敏,适用于大鼠血浆中Dox的含量测定,同时为Dox及CS-Dox大鼠药代动力学研究提供了可靠的分析方法。
Objective:To establish a new method for determining the content of doxorubicin(Dox) in rat plasma. Methods:Dox in rat plasma was determined by fluorescence spectrophotometry. A rapid, simple and sensitive method for the quantitative determination of chondroitin sulfate-doxorubicin in rat plasma was established. Results:A good positive linear relationship was shown in the mass concentration range of Dox 0.1-15.0 μg/mL. The regression equation was Y=16.944 X+0.6796,R2 = 0.9997. The RSDs of Dox and CS-Dox within-day and between-day were less than 2.5%, and the extraction recovery of Dox and CS-Dox was greater than 84%. The results indicated that this method has high recovery rate. Conclusion:The assay is inexpensive, simple, precise, sensitive and suitable for the determination of doxorubicin in rat plasma. At the same time, it provides a reliable analytical method for the pharmacokinetic study of Dox and CS-Dox in rats.
