巴什拜羊H-FABP基因遗传多态性及其与生长性状的相关分析

摘要
旨在运用分子生物学手段研究H-FABP基因遗传多态性与巴什拜羊生长性状的关系,为今后用分子标记辅助育种方法提高巴什拜羊生长性状提供科学依据。以300只巴什拜周岁母羊为研究对象,选取H-FABP基因外显子2和外显子3的部分片段,采用PCR-SSCP和DNA测序等技术检测其遗传多态性,并与体重、体长、体高等性状进行关联分析,研究H-FABP基因的多态性与巴什拜羊生长性状的关系。结果表明,H-FABP-P_2基因座外显子3无多态性;H-FABP-P_1基因座外显子2存在AA和AG2种基因型,测序结果显示在H-FABP基因外显子2的938bp处发生了A→G碱基突变,为同义突变。经关联分析发现,AG基因型个体的胸围和体重显著高于AA基因型个体(P<0.05)。因此,巴什拜羊H-FABP基因外显子2上的点突变可能是影响巴什拜羊生长性状的重要位点。
        In this study, in order to provide scientific basis for improving growth traits of Bashibai sheep by molecular marker assisted breeding methods, the relationship between the genetic polymorphisms of H-FABP gene and the growth traits was studied by molecular biology methods. In this experiment, 300 Bashibai yearling ewes were taken to select part of segments of H-FABP gene exon 2 and exon 3 and study their genetic polymorphisms by PCR-SSCP and DNA sequencing. Its association with body weight, body length, and body height was then explored to study the relationship between H-FABP gene polymorphism and these indexes of Bashibai sheep. Results show there was no polymorphism in H-FABP-P_2 gene locus(exon3); there was polymorphism in H-FABP-P_1 Gene locus(exon 2), which had 2 genotypes: AA and AG. Sequencing results showed that there was a base mutation A →G in location 938 bp of H-FABP gene exon 2, which was synonymous mutation. Correlation analysis found that the individual weight and heart girth of AG genotype were much higher than that of AA genotype(P<0.05). The point mutation on exon 2 of H-FABP gene in Bashibai sheep may be an important site affecting the growth traits of Bashibai sheep.
