水溶性三芳基硼化合物用于细胞线粒体内H_2S检测的研究

摘要
在生物体内,硫化氢是一种多功能信号分子,参与许多重要的生理过程。鉴于硫化氢分子的重要性,我们设计并合成了一系列基于三芳基硼荧光团的硫化氢探针TAB-1-Cu~(2+)、TAB-2-Cu~(2+)和TAB-3-Cu~(2+)。探针与硫化氢反应后,原本被猝灭的可发光重新显现,实现高效、灵敏、准确的硫化氢荧光检测。硫化氢分子还可导致探针分子发生有限的聚集,大大增强了其稳定性和对环境的耐受性。此外,细胞实验表明,TAB-2-Cu~(2+)具有良好的跨膜性、较低的细胞毒性、线粒体定位性,因此可以对线粒体的硫化氢实时成像。使用TAB-2-Cu~(2+),可以通过双光子显微镜、荧光寿命成像技术对NIH/3T3细胞中的硫化氢分布成像。
Hydrogen sulfide(H_2S) is a multifunctional signaling molecule that participates in many important biological processes. Herein, by functionalizing triarylboron with cyclen and diphenylamine, we designed and synthesized TAB-1, TAB-2 and TAB-3 for H_2S recongnization. TAB-2 possessed excellent membrane permeability and could effectively complex with Cu~(2+). The complex TAB-2-Cu~(2+) could selectively detect H_2S with an instant response and mitochondria targeting effect. TAB-2-Cu~(2+) was also successfully applied for the mitochondria H_2S imaging in NIH/3T3 fibroblast cells by two-photon microscopy and fluorescence lifetime imaging microscopy.
