基于PCR-DGGE技术分析东南沿海城市水产品的细菌群落结构

摘要
微生物群落在食品质量和安全中起着重要的作用,被认为是导致食品腐败和食源性疾病的主要原因。基于PCR-DGGE的变性梯度凝胶电泳和16SrRNA基因克隆文库进行分析,对东南沿海的五个城市(上海、杭州、福建、宁波、福州)水产品进行检测。结果表明:变形菌门、厚壁菌门在五个城市中占主要地位,五个城市的水产品中都检测出弧菌属、巨球菌属。上海检测出不动杆菌属最多;其中发光杆菌属在厦门、福州作被检测;肠球菌属、气单胞菌属分别在杭州、宁波被大量检测出。香农-威纳指数表明细菌多样性具有显著差异,其中宁波最为明显。应用该方法在细菌群落的分析可以帮助我们更好地了解水产品中细菌的多样性和分布,为水产品中风险评估提供了参考。
Microbial community plays a prominent role in food quality and safety,which was regarded as the major cause for food spoilage and foodborne disease,.PCR-based denaturing gradient gel electrophoresis(DGGE) and 16 SrRNA gene library analysis were performed to analyze the bacterial community of aquaticproduct infive cities(Shanghai,Hangzhou,Fujian,Ningbo,Fuzhou) in the southeast coast.The results of DGGE profiles showed that Proteobacteria,Firmicutes dominated in five cities,Vibrio sp、 Enterococcus sp existed in five cities.Shanghai detected Acinetobacter sp most;where in Photobacteriumsp in Xiamen.,Fuzhou,;Enterococcus sp 、 Aeromonas spdetectedin Hangzhou,Ningbo respectively.Shannon-Wiener index showed significant difference in bacterial diversity,which is most evident in Ningbo.In conclusion,this study provides an effective tool foranalysis of bacterial community,might help us better understand the diversity and distribution of bacteria in aquaticproduct,This study provided references for the risk assessment of Vibrio parahaemolyticus.
