姬玉露不定芽诱导及高频离体再生体系的建立

摘要
以姬玉露叶片为外植体,研究不同激素配比对愈伤组织诱导、再生和生根的影响,建立姬玉露高频离体再生体系。结果表明:愈伤诱导最佳配方MS+6-BA1.0mg/L+NAA0.1mg/L。芽诱导最佳培养基配方:MS+6-BA0.8mg/L+TDZ0.01mg/L+NAA0.3mg/L。增殖最佳培养基配方:MS+ZT0.8mg/L+NAA0.1mg/L,增殖系数达到10.04。生根最佳培养基配方:1/2 MS+NAA0.2mg/L。
With the leaf of Haworthia cooperi var.truncate as the explants,the influence of different ratio of hormones on the callus induction,regeneration and rooting was studied and a high frequency in vitro regeneration system was established.The results showed that:with the 0.1%HgCl2 and processing 8 min has the best effect and the rate of survival is 76.23%.The optimum medium for callus induction is MS + 6-BA1.0mg/L + NAA0.1mg/L The adventitious bud induction is MS + 6-BA0.8mg/L+ TDZ0.01mg/L + NAA0.3mg/L The medium for propagation of adventitious bud is MS + ZT0.8mg/L +NAA0.1mg/L,and the propagation factor is 10.04.The optimum for rooting is 1/2 MS + NAA0.2mg/L
