摘要
棉花纤维是由棉花胚珠外表皮细胞在短时间内分化发育而形成的单细胞结构。极性脂质是纤维发育过程中细胞膜膨胀与细胞快速伸长所必需的,但目前对纤维细胞中脂类的运输及其调控纤维伸长发育的机制并不清楚。本研究报导了功能性蛋白GhLTPG1在棉花纤维伸长发育过程中的作用。GhLIPG1是1个GPI锚定的脂质转运蛋白,是实现脂类运输的候选蛋白。GhLTPG1为纤维特异表达基因,在纤维及胚珠外表皮层细胞中表达十分丰富。GhLIPG1蛋白在烟草与拟南芥中均定位在细胞膜与核膜上,在拟南芥中还具有叶表皮毛与种子外表皮定位的特点。生化分析表明GhLTPG1在体外能够特异性地结合单磷酸磷脂酰肌醇(PIP),而对双磷酸及三磷酸磷脂酰肌醇无亲和力,并且可以在体内把单磷酸磷脂酰肌醇从合成位点运输到质膜。在拟南芥中过表达GhLTPG1可以使腺毛数目增加,在棉花中干扰GhLTPG1表达后会导致纤维长度变短,使胚珠中PI及总极性脂的含量显著降低,以及导致一系列与纤维伸长发育相关基因的表达受抑制。这些结果表明GhLTPG1通过调控单磷酸磷脂酰肌醇的转运来调控棉花纤维的伸长。
Cotton fibers are single cells that rapidly derived from the ovule epidermis in a short term.Polar lipids are required for the vigorous enlargement of cell membrane and the quick growth of fiber cells.However,the mechanism how the lipids are transported from the ovules into the developing fibers and their effects on fiber elongation remains unknown.Here,we reported the functional characterization of GhLTPG1 during cotton fiber elongation.GhLTPG1 is a GPI-anchored lipid transport protein,and it is the candidate protein involved in lipid transportation.GhLTPG1 is a fiber specific expression gene,abundantly expressed in elongating cotton fibers and outer integument of the ovules.GhLTPG1 dominantly located on the cell membrane and nuclear membrane in both tobacco and Arabidopsis,and showed a leaf trichome and seed coat localization in transgenic Arabidopsis.Biochemical analysis showed that GhLTPG1 can specifically binds to phosphatidylinositol-monophosphate(PIP) in vitro rather than di-or tri-phosphate phosphatidylinositol,and transported PtdlnsPs from the synthesis places to the plasma membranes in vivo.Expression of GhLTPG1 in Arabidopsis caused an increased number of trichomes,and fibers in GhLTPG1-knockdown cotton plants exhibited significantly reduced length,decreased polar lipid content,and repression of fiber elongation-related genes expression.These results suggested that GhLTPG1 protein regulates the cotton fiber elongation through mediating the transport of phosphatidylinositol monophosphates.
