The inside back cover picture shows the ability of dendrimeric amino- and, in a much more efficient way, guanidinoneomycin-PAMAM vehicles to promote the cellular uptake of high-molecular-weight biologicals, such as enzymes or proteins. These conjugates have been synthetized by tethering neomycin and guanidinoneomycin to PAMAM dendrimers of generations 2 and 4 through isothiocyanate-amine click chemistry. The mechanism of entry depends on the presence of heparan sulfate proteoglycans on the cell surface, as no uptake has been observed in mutant cells lacking heparan sulfate.
