Effect of enzyme load and catalyst particle size on the diffusional restrictions in reactions of synthesis and hydrolysis catalyzed by α-chymotrypsin immobilized into glyoxal-agarose
α-Chymotrypsin was immobilized in glyoxal agarose. Hydrolysis and synthesis reactions were modelled using Michaelis-Menten kinetics. Km and Vm were determined on different support sizes and enzyme loads. Internal diffusional restrictions were evaluated in hydrolysis and synthesis. Maximum size and load to avoid diffusional restrictions were determined.
