Sensitive and selective detection of miR-21 is vital to the clinical diagnosis for breast cancer.

A target-triggered triple isothermal cascade amplification strategy has been newly developed.

A tri-functional DNA template contains target recognition, analogue amplification and DNAzyme production parts.

Polymerase-nicking enzyme, 8-17 DNAzyme and HRP-mimicking DNAzyme were employed in the cascade reaction.

Attomolar sensitivity with excellent selectivity for the quantification of miR-21 is realized.
