Around 7000 variants of an 伪-lass="smallcaps">l-arabinofuranosidase were prepared by error prone PCR.
The four selected variants showed the same aminoacidic substitutions F435Y/Y446F.
The variant F435Y/Y446F was overproduced in Pichia pastoris and purified and characterized.
The variant F435Y/Y446F showed a kcat 3 fold higher than the wild-type towards p-NPA.
The variant is able to hydrolyze larch arabinogalactan not recognized by the wild type.
