Raman and SERS Spectroscopy of D96N Mutant Bacteriorhodopsin

Effect of silver nanoparticles (AgNPs) on the D96N mutant form of bacteriorhodopsin (BRh-D96N) contained in fragments of purple membranes (PMs) of bacteria Halobacterium salinarum has been analyzed. It has been found that interaction of AgNPs with BRh-D96N in SERS-active spots leads to “freezing” of the protein photocycle. The AgNPs change and then fix the state of BRh-D96N in SERS-active spots. Outside of SERS-active spots, the effect of AgNPs is weaker and is expressed in deceleration of the M-form decay. Thus, the fundamental difference in the influence of AgNPs on the mutant BRh-D96N in comparison with wild-type BRh was found.
