BCL-2 proteins control the intrinsic or mitochondrial pathway of apoptosis. Defective apoptosis is a hallmark of cancer. BH3-mimetics inhibit pro-survival BCL-2 proteins to induce cancer cell death. ABT-199/venetoclax is approved for treatment of chronic lymphocytic leukaemia.
