Evidence that DNA adducts in some human kidney tumours in France are related to ochratoxin A

Ochratoxin A (OTA), a natural contaminant of mouldy food and feed, is suspected of being one of the etiological agents responsible for Balkan endemic nephropathy (BEN) and associated urinary tract tumours. OTA induces renal adenomas in rodent. In order to provide if OTA, even in France, could be implicated in the etiology of some human urinary tract cancer, kidney tumours and peritumorous tissues from 20 patients were obtained from surgery. Since the first step in the tumour formation is the alteration of DNA, we have analysed DNA adduct and compare them to DNA adduct observed in OTA-treated rodent. Moreover, OTA in blood of the patient have been measured. Several DNA adducts were detected in all peritumourous tissues. The total adduct levels ranged between 5 to 50 adducts/109 nucleotides. Lower amounts are observed in tumoral tissue of the same individuals. DNA adduct patterns differed from one individual to the other, indicating different kind of contaminants. Several DNA adducts with the same Rf values as those obtained from OTA treated rodent were detected in some patient, but not in all. These results provide evidence of the possible role of OTA in some kidney tumours.
