Visualized genotyping of multiple targets was achieved by detecting multiplexed LAMP amplicons with series target-specific invasive reactions coupled with hybridization-induced assembly of gold nanoparticles. The key steps of the method include sequence amplifier to amplify the multiple targets, allele identifier to recognize a targeted base in the multiplexed amplicons, and signal generator to yield red color from disconnected gold nanoparticles. Since sequence amplifier and allele identifier occurs at a constant temperature, the method only needs a simple temperature controller, much suitable for source-limited regions to assist with gene-guided personalized medicine.
