Located at the turning point of hemagglutinin2 (HA2) conformational change during membrane fusion of influenza virus, a conserved stem fragment with both B- and T-cell epitopes was identified.
The immunization of conserved stem fragment could induce strong humoral immune response and cross-clade neutralizing antisera against strains from H3 subtype in group 2.
The cross-clade neutralizing antisera possessed neutralizing activities potentially by blocking conformational change at the turning point of HA stem where exposed for limit periods of time during membrane fusion.
