A biological method for producing crotonic acid through reversal β-oxidation (r-BOX) was proposed.

YdiI, an Escherichia coli thioesterase, was shown to convert trans-enoyl-CoAs to α,β-unsaturated carboxylic acids (α,β-UCAs).

Production of medium chain α,β-UCAs through r-BOX platform was demonstrated.

Superiority of r-BOX over fatty acid biosynthesis pathway was validated through the use of genome scale model.
