Using containerless methods to develop amorphous pharmaceuticals

Containerless methods completely eliminate extrinsic heterogeneous nucleation and they can be used to make samples of phase-pure amorphous drugs even from reluctant glass formers. Acoustic levitation was shown to be suitable for containleress processing of supersaturated solutions and supercooled melts formed from drug and drug-binder samples. The method extended the range of drug compositions that could be made into amorphous forms. In-situ structure measurements using high energy X-ray diffraction were made during the amorphization process, these measurements provide a valuable tool to follow the process of amorphization. Samples made using containerless processing and spray drying have similar structures. This finding suggests that acoustic levitation can be used in development and characterization of materials that can be scaled up using spray drying.
