Seamless deletion of a large DNA fragment in the taxol-producing fungus Pestalotiopsis microspora

A genome reduction method was developed and tested in Pestalotiopsis microspora. Gateway/overlap extension PCR were applied to avoid tedious plasmid operations. Two adjacent DNA sequences were sequentially removed in a single transformation step. The deletion cassette can be transformed by ATMT with high efficiency. A large genomic region was deleted with one construct, with no residual junk DNA.
