Target analysis of total thyroid hormones in serum was performed by LC-QTOF-MS. Challenging determination due to the low concentrations in a very complex matrix. Endogenous serum components cause remarkable ionization suppression effects. Deproteinization and mixed-mode SPE led to significant reduction of matrix effect. Retrospective analyses allowed detection of co-eluting lysophosphatidylcholines.
