Photophysical studies on a photoactive yellow protein fluorophore analog with the 4-Hydroxy group replaced by 4-Dimethylamino group

A highly polarity sensitive PYP Fluorophore analog – pDMACT – has been prepared, exhibiting fluorescence Stokes shifts comparable to PRODAN. Unlike previous analogs, here the p-phenolate group is replaced – instead of the carbonyl end group – by p-N,N-dimethylaminophenyl group. pDMACT is thus a prototype for new fluorescent labels that can be attached to a biomolecule by the thioester linkage. Its structure promotes strong intramolecular charge transfer, which plays a major role in its excited state relaxation in bulk solvents.
