Chromatin caused precipitation and loss of IgG during sample equilibration.

It also depressed cation exchange capacity and IgG recovery.

Advance chromatin extraction suspended these limitations.

HCP was reduced to <1000 ppm, DNA to <1 ppb, and aggregates to <2.5%.

A single polishing step reduced HCP to 15 ppm and aggregates to <0.1%.
