The GluN3A NTD constitutes a crucial regulatory determinant of GluN1/GluN3A receptor function.

This is attributed to the selective loss of agonist-evoked auto-inhibition caused by glycine-binding to the GluN1 subunit.

The GluN3A NTD subunit interface and NTD LBD linker participate in regulating the efficacy of glycine at GluN1/GluN3A receptors.
