Replacement of two amino acids of 9R-dioxygenase-allene oxide synthase of Aspergillus niger inverts the chirality of the hydroperoxide and the allene oxide

A set of dioxygenase-P450 fusion enzymes of Aspergilli remains to be characterized.

Expression of one of these enzymes revealed a 9R-dioxygenase-allene oxide synthase.

Sequence analysis showed homology to fungal 9S-dioxygenase-allene oxide synthases.

Replacements in the 9R-dioxygenase domain could explain the catalytic differences.

We conclude that 9R- and 9S- dioxygenase-allene oxide synthases are closely related.
