We proceeded to express a small chitin synthase from Rhizopus oryzae in E. coli. The heterologous enzyme was active and synthesized chitin partially microfibrilar. The enzyme was partially purified when inclusion bodies were solubilized with urea. Kinetic properties were unusual indicating the requirement of transmembrane elements. This approach may lead the to further isolation of chitin synthases to determine their mechanism.
