Polκ extends DNA from a major bulky adduct with high fidelity by kinetic analysis. Polκ accommodates bulky benzo[a]pyrene (BP) adducted DNA in a B-form. The bulky BP ring in a 5′ orientation protects nascent DNA in the minor groove side. Minor groove protection acts as base pairing validation for accurate replication.
