Deubiquitinating enzyme USP21 was identified to interact with NANOG by yeast two hybrid screening for DUB enzymes. NANOG can directly bind to USP21. Overexpression of wild-type USP21, but not USP21 (C221S), leads to increased expression level of NANOG. K48-specific, but not K63-specific, polyubiquitin chains of NANOG are decreased by USP21.
