The use of myeloablation and bone marrow transplantation allowed the generation of chimeric mice to study the role resident microglia and infiltrated bone marrow-derived macrophages (BMDM) separately in the brain.

We here review the advent of this methodology and compare the different strategies used to perform myeloablation.

We discuss the roles of microglia and BMDM in neurological disorders.
