Codon deoptimization (CD) results in reduced LCMV GP expression levels. A rLCMV encoding a CD GP (rLCMV/CD) shows WT-like growth properties in vitro. rLCMV/CD is highly attenuated, as compared to LCMV WT, in vivo. rLCMV/CD induces complete protection against a lethal challenge with rLCMV/WT. A GP CD-based strategy can be implement for the development of arenavirus vaccines.
