The mechanistic trigger of cell senescence from a condition of autophagy deficiency is proposed. Betulinic Acid (BA) physically interacts and disorganizes membranes, causing membrane leakage in membrane mimics and in intracellular organelles. Parallel damage in the membranes of mitochondria and lysosome causes lipofuscinogenesis, genomic instability and cell senescence. Senescence is the long-term consequence of treating cells with BA.
