Silica encapsulated Au with mercaptobenzoic acid core-shell nanoparticles (Au-MBA@SiO2) was developed for the SERS-based immunoassay. The silica shell was employed to protect SERS labels, and to block the aggregation of Au nanoparticles and the nonspecific bingding. MBA molecules played dual roles as SERS labels and to facilitate the formation of silica shell The detection sensitivity was improved by about 1– 2 orders of magnitude by comparing to the traditional approach based on naked Au-MBA nanoparticles. This kind of labels embedded core-shell nanoparticles could be developed as the versatile nanotags for the bioanalysis and bioimaging.
