Eukaryotic mitochondria have a full set of discrete enzymes for de novo synthesis of fatty acids. Mitochondrial fatty acid synthesis proceeds in an ACP-dependent manner. Failure leads to respiratory deficiency in yeast and embryonic lethality in mammals. Lipoic acid metabolism and mitochondrial fatty acid synthesis are tightly interlinked. Human mitochondrial ketoreductase (KAR) of mitochondrial fatty acid synthesis links this process to mitochondrial β-oxidation.
