TGF-β1 exerts apoptotic, necrotic, and mitoinhibitory effects in L1210 leukemic cells. Apoptosis is induced at low TGF-β1 concentrations (0.001 - 0.1 ng/ml), whereas mitoinhibition at concentrations exceeding 1 ng/ml. Necrosis is probably a secondary to apoptosis form of death, since TGF-β1 is not considered as a cytotoxic agent. Apoptosis is manifested by cell shrinkage, chromatin condensation and margination, nuclear pycnosis and fragmentation. Characteristic “DNA ladder” resulting from its internucleosomal fragmentation (multiples of 180 - 200 bp) was visualised by gel electrophoresis. Flow cytometry analysis demonstrated that apoptosis affects mainly cells in G1 phase. The induction of apoptosis by TGF-β1 depends on the supply of growth and trophic factors present in fetal calf serum (FCS). FCS deprivation and bovine serum albumin supplementation enhances cell susceptibility to TGF-β1-induced apoptosis.
