We have evaluated the performances of PneumoGenius real-time PCR that combines Pneumocystis amplification and the detection of DHPS mutations. PneumoGenius real-time PCR has shown comparable performances to an ‘in house’ real-time PCR and another commercial quantitative real-time PCR assay, the Bio-Evolution Pneumocystis jirovecii real-time PCR, in discriminating Pneumocystis pneumonia and colonization. A 100% agreement was obtained between PneumoGenius® PCR and the “in house” PCR and sequencing method for DHPS mutation identification We observed a low rate of P. jirovecii harboring DHPS mutations (4.5%) in our hospital population.
