SH2B1 fine-tunes global and regional chromatin structures and promotes myogenesis. SH2B1 regulates epigenetic modification and enhances MyoD transcriptional activity to control expressions of myogenic genes. A novel distal E-box (− 6.5 k) MyoD binding site for MYOG gene was identified. SH2B1 binds to histone H1 and is required for the removal of histone H1 at IGF2 and MYOG promoters. SH2B1 is a novel target of MyoD.
