P106 Photo-manipulation of vasodilation with a novel photo-controllable nitric oxide releaser based on N-nitrosoaminophenol

Nitric oxide (NO) is synthesized in the human body and plays main roles in some physiological event, such as vasodilation, neurotransmission, and immune response. Due to its instability in physiological conditions, NO releasers, which can store and release NO, had been utilized for the research, so far. Among them, photo-controllable NO releasers are expected as precisely controllable chemical tools or agents for photodynamic therapy. Although many photo-controllable NO releasers had been reported, there are few report of visible-light controllable NO releaser which could photo-control physiological events ex vivo. We designed and synthesized a novel visible light controllable NO releaser, named NOBL-1, composed of N-nitrosoaminophenol as an NO releasing moiety and BODIPY as a visible light harvesting moiety. NO release from NOBL-1 induced by blue light was confirmed by means of ESR spin trapping method with iron-dithiocarbamate complex. Furthermore, it was confirmed that the NO release was under control in temporal and spatial precision in vitro and in cell by using a fluorogenic NO probe, DAR-4M. Finally, we achieved vasodilation control of rat aorta slice with NOBL-1 and the triggering light in highly temporal manner. NOBL-1 is expected to be a useful chemical tool for NO research, and it may also have potential for phototherapy.
