Le CL est oxydé, en présence de cholestérol oxydase, en cholesténone avec libération de peroxyde d’hydrogène ; ce dernier oxyde l’amino-4-antipyrine, en présence de phénol et de peroxydase en une quinone imine colorée dont l’intensité de la coloration évaluée à 510 nm est proportionnelle à la quantité de CL.
Une parfaite linéarité entre l’absorbance et la concentration du CL a été notée avec une droite d’étalonnage du type Y = aX. Des coefficients de précision et d’exactitude inférieurs à 5 % sont enregistrés jusqu’à une concentration de 4,1 mmol/L. Les résultats de l’étude de la répétabilité et de la reproductibilité sont très satisfaisants avec des CV < 5 % et sans différence significative entre les moyennes pour les sérums titrés. Une bonne exactitude évaluée par les procédés de mélange et de dilution a été notée. Une parfaite corrélation avec les résultats du kit commercialisé DiaSys est retrouvée.
La technique enzymatique colorimétrique pour le dosage du CL est précise, exacte et rapide. Elle peut être introduite en routine quotidienne pour la détermination du CL. Son adaptation au dosage du CL des HDL constitue un second sujet d’étude.
Free cholesterol (FC) is the marker of atherosclerosis less required in spite of its description in the atherosclerotic plaques and the vessels of the coronarian subjects. The objective of this work is to evaluate and validate a FC enzymatic method in order to apply it in daily practice in the exploration of the lipid parameters.
FC is oxidized, in the presence of cholesterol oxidase, in cholestene-3-one with hydrogen peroxide release; this last product oxidizes 4-aminoantipyrine, in the presence of phenol and peroxidase in a coloured quinoneimine evaluated at 510 nm.
A perfect linearity between the absorbance and the concentration of FC was noted with a calibration curve of the type Y = aX. Precision and exactitude Coefficients were lower than 5 % until a concentration of 4,1 mmol/L. Repeatability and reproducibility are very satisfactory (< 5 % ) and without significant difference between the averages for the titrated serums. A good exactitude evaluated by the processes of mixture and dilution was noted. A perfect correlation with the results of the kit marketed DiaSys is found.
The colorimetric enzymatic technique for the determination of free cholesterol is precise, exact and fast. It can be introduced in daily routine for the determination of FC. Its adaptation to the determination of the FC of the HDL constitutes a second subject of study.
