We report on a method for chemiluminescence determination of the caspase-1 using peptide and luminol labeled gold nanoparticles as a chemiluminescence probe and magnetic beads as immobilization matrix. The chemiluminescence reagent luminol and caspase-1 specific peptide labeled gold nanoparticles were firstly prepared. Then they were conjugated to the carboxyl terminated magnetic beads to form functionalized magnetic beads. The site YVAD contained on functionalized magnetic beads can be cleaved by caspase-1. On addition of sample caspase-1, it will cleave peptide into two parts. One part attached to gold nanoparticles can dissociate from functionalized magnetic beads and can be used for chemiluminescence detection. Under optimal conditions, caspase-1 can be detected in the concentration ranged from 0.01 ng mL−1 to 30 ng mL−1, and the limit of detection was 0.003 ng mL−1. The reproducibility and selectivity of the developed method were also investigated. In addition, the real samples were assayed by using the developed method, and the recovery was 96.0–102.0%. This method holds promise as a widely applicable general platform for proteins detection.
